bacteriophages are mixed with bacteria and then the bacteria with attached phages are mixed with melted agar

melted agar is poured in a very thin layer over solidified nutrient agar

as the viruses replicate, they kill the hos cell and are released into the adjacent agar (can infect adjacent bacteria and kill them too)

result is a transparent spot in a cloudy lawn of growing bacteria( this is called a plaque) and corresponds to one bacteriophage
